AJB Primer Notes & Protocols in the Plant Sciences is an online-only section of the American Journal of Botany (AJB) intended to promote rapid dissemination of protocols used in genetic analyses of plants. Subjects appropriate for this section include: (1) marker notes providing primer sequences for microsatellite or other markers in particular taxa, and (2) protocol notes describing new methods for isolating, visualizing, or scoring genetic markers. Protocol notes are expected to provide evidence that the new protocol increases the accuracy or reproducibility of marker scoring, decreases the time or expense of marker scoring, or provides the ability to score new classes of markers.

For primer notes, the intent is to demonstrate the usefulness of newly developed primers for subsequent, more detailed studies.  Sampling distribution and numbers will depend on the life history characteristics of the organism under study, but sample sizes should be adequate to support the genetic parameters presented.  For example, sample sizes are expected to be greater for abundant and widely distributed species than for rare species.  Whenever possible, the use of data from multiple populations is encouraged to capture a wider range of potential variation.  Although most primers are expected to exhibit polymorphism, monomorphic primers are also acceptable because they may be useful in other populations or related taxa.  Given the limited sampling and biological characteristics of some species (small population sizes, recent colonization, admixture, clonality, etc.), tests of linkage disequilibrium and Hardy-Weinberg equilibrium are not required for primer notes articles but are instead reserved for later in-depth population surveys.
